3 miRNA panel inhibits OECM-1 invasion and migration. (a,b) Cell migration using a wound healing assay shows a significant reduction in the migratory ability of OECM-1 cells transfected with miR-143/miR-145/miR-140 mimics as compared to those treated with SC. (c,d) A transwell assay for cell invasion showed a significant reduction in the proportion of invaded cells when transfected with the combination of 3 miRNA mimics as opposed to SC. (Scale: 59 μm). (e) Phase contrast images show clear visual reduction in the proportion of mesenchymal cells after transfecting OECM-1 cells with the combination of 3 mimics. On the contrary, a greater proportion of mesenchymal cells can be seen in the SC population. (f) Representative bar graphs of qRT-PCR-based expression analysis of E-cadherin and N-cadherin in OECM-1 cells after transfection depict relative upregulation of E-cadherin and downregulation of N-cadherin. The expression levels of genes were estimated using real-time PCR. The data were normalised with β-actin values, and the relative gene expression levels were analysed using the ddCt method.
