Macrophages distinguish between “self” or “non-self” by binding to SIRPα transmembrane protein on macrophage to form the CD47/SIRPα signaling complex. A CD47 expressed on cancer cell membrane binds to SIRPɑ on macrophage cell membrane to activate the “Don’t eat me” signal and block macrophage phagocytosis of cancer cells.
