Exogenously added miR29b strongly increases MM plasma cell lysis by bone marrow-derived cytotoxic NY-ESO-1157–165-specific CD8+ T cells. In this cytotoxicity assay, effectors were CD8+ T cells (from bone marrow of 5 MM patients) that had been expanded in vitro in the presence of autologous antigen-presenting cells pulsed with NY-ESO-1157–165 peptide. Targets were untransfected, untreated U266 cells or U266 cells transfected with 10 µM miRNA mimic negative control (miR-NC), 10 µM miR29b mimic (miR29b), or treated with 1 µM decitabine for 72 h. *P < .05, ***P ≤ .001.
