A schematic picture of synNotch-CAR (a) and iCAR (b). SynNotch-CARs require T cells to sense two antigens to activate. SynNotch receptors are engineered to sense a target antigen on the surface of tumor cells and induce the expression of a CAR specific to a second tumor antigen. iCAR-T cells express two CARs on the same T cells, including a typical tumor-antigen-specific CAR and an iCAR.
